The proteolytic system involving calpains.

چکیده
Autophagosomes were isolated from perfused livers on Percoll gradients after the administration of cathepsin inhibitors (Kominami e t aZ., 1983). Cycloheximide (1.8 X lo-' M ) or vinblastine ( 1 X M ) were administered either simultaneously or 60 min after the protease inhibitors. ( a ) Acid phosphatase accumulation in autophagosomes (units/g of liver); ( b ) protein accumulation in autophagosomes (mg/g of liver). 0----0, Control; A--.-n, cycloheximide at zero time; A---A, cycloheximide at 60 min; m. , vinblastine at 60 min.

Intracellular proteolytic systems in alcohol-induced tissue injury.
The body constantly produces proteins and degrades proteins that are no longer needed or are defective. The process of protein breakdown, called proteolysis, is essential to cell survival. Numerous proteolytic systems exist in mammalian cells, the most important of which are the lysosomes, the ubiquitin-proteasome pathway, and enzymes called calpains. Lysosomes are small cell components that co...

The calpains (E.C. 3.4.22.17) and calpastatin constitute an ubiquitous, intracellular, Ca2+dependent protease/inhibitor system. This system has been implicated as a principal regulator of myofibrillar protein degradation in both ante-mortem and postmortem muscle. Although proteolytic activity of the calpains is primarily controlled through interaction of calpain and calpastatin, evidence for an...
متن کاملRegulation and physiological roles of the calpain system in muscular disorders
Calpains, a family of Ca(2+)-dependent cytosolic cysteine proteases, can modulate their substrates' structure and function through limited proteolytic activity. In the human genome, there are 15 calpain genes. The most-studied calpains, referred to as conventional calpains, are ubiquitous. While genetic studies in mice have improved our understanding about the conventional calpains' physiologic...
متن کاملEffects of serum deprivation on expression of proteolytic-related genes in chick myotube cultures.
We previously reported that serum deprivation stimulates myofibrillar proteolysis in chick myotubes. In the present study, we examined the effect of serum deprivation on expression of the proteolytic-related genes (ubiquitin, proteasome, calpains, and cathepsin B) by real-time PCR of cDNA in chick myotubes. Myotubes were incubated with serum-free medium for 24 h. Ubiquitin and proteasome subuni...
متن کاملThe inhibitory role of sympathetic nervous system in the Ca2+-dependent proteolysis of skeletal muscle.
Mammalian cells contain several proteolytic systems to carry out the degradative processes and complex regulatory mechanisms to prevent excessive protein breakdown. Among these systems, the Ca2+-activated proteolytic system involves the cysteine proteases denoted calpains, and their inhibitor, calpastatin. Despite the rapid progress in molecular research on calpains and calpastatin, the physiol...
